Reactive Oxygen Species (ROS) Detection Assays

Reactive oxygen species (ROS) are chemically reactive molecules such as superoxide, hydrogen peroxide and hydroxyl radicals which are continually formed as natural byproducts of the normal aerobic metabolism. They play a critical role in cellular physiopathology and are implicated for example in cell proliferation, cell signaling pathways, homeostasis, apoptosis, thyroid hormon synthesis, as well as oxidative defense mechanisms (e.g. killing of bacteria). ROS generation is normally counterbalanced by the action of antioxidant enzymes and other redox molecules such as glutathione (GSH). In several disorders such as cancer or neurodegenerative and cardiovascular diseases, the redox balance is disturbed and excessive amounts of ROS can can lead to cellular injury by damaging cellular structures, proteins, membrane lipids and DNA, which is known as oxidative stress.
Thus, having a sensitive method for detection of intracellular ROS is very important for detecting the disturbed redox balance in pathological cells as well as cells that have been exposed to different genotoxic agents (chemicals, irradiation, xenobiotics, etc.).

Reactive Oxygen Species (ROS) Detection Kit

PromoKine’s Reactive Oxygen Species (ROS) Detection Kit provides a simple, specific and highly sensitive method for the real-time measurement of ROS levels in living cells or antioxidant activity of test compounds. It utilizes H2DCFDA, a cell-permeable fluorogenic probe, compatible with phenol red, FBS and BSA to detect hydroxyl, peroxyl, or other reactive oxygen species in live cells. Upon the cell entry, H2DCFDA is modified by cellular esterases to form a non-fluorescent H2DCF which is oxidized by intracellular ROS to yield a highly fluorescent product that can be detected by using FACS, microplate reader, or fluorescence microscope (Ex/Em 495/529 nm). The fluorescence intensity is proportional to the ROS levels.

Hydrogen Peroxide Detection Kits

Hydrogen Peroxide (H2O2) is a reactive oxygen species and by-product of oxygen metabolism. It is generated from superoxide (which is produced by mitochondria and NADPH oxidases) by the enzyme Superoxide Dismutase (SOD) which thus prevents accumulation of superoxide that can e.g. damage and inactivate proteins causing oxidative stress. H2O2 serves as a key regulator for a number of oxidative stress related pathways and is linked to asthma, inflammatory arthritis, atherosclerosis, diabetic vasculopathy, osteoporosis, Down’s syndrome, immune system diseases and a number of neurodegenerative disorders.

PromoKine's Hydrogen Peroxide Assay Kit is a highly sensitive, simple, direct and HTS-ready colorimetric and fluorometric assay for measuring H2O2 in various biological samples. In the presence of Horse Radish Peroxidase (HRP), the OxiRed Probe reacts with H2O2 resulting in a product that can be quantified colorimetrically (λmax = 570 nm) as well as fluorometrically (Ex/Em=535/587 nm). The kit can perform 200 reactions by using the fluorometric method or 100 reactions by using the colorimetric method. The detection limit can be as low as 2 pmol per assay (or 40 nM concentration) of H2O2 in the more sensitive fluorometric assay.
