Nitric Oxide (NO) and Nitric Oxide Synthase (NOS) Detection Kits

Nitric oxide (NO) which is produced by nitric oxide synthase (NOS) is also classified as reactive oxygen species (ROS), but sometimes also as reactive nitrogen species (RNS). It plays a key role in neurotransmission, vascular regulation, immune responses, cell communication, and apoptosis. NO is produced by immune cells, including macrophages, as a part of body’s defense mechanism during immunological responses and oxidative stress conditions. However, dysregulation or chronic overproduction of NO is considered to be implicated in the pathogenesis of many disorders including neurodegenerative, inflammatory, vascular  and autoimmune diseases, diabetes, renal ischemia, cancer and artherosclerosis.

PromoKine's Fluorometric Nitric Oxide Kit provides an accurate and convenient tool for total nitrate/nitrite concentration determination in a simple two-step process that can be used as a quantitative measure of NO production. The first step is the conversion of nitrate to nitrite utilizing nitrate reductase. The second step involves the addition of the fluorometric reagent DAN followed by NaOH, which converts nitrite into a fluorescent compound. Measurement of the fluorescent compound reaction product accurately determines the total nitric oxide production. The Fluorometric Assay Kit has been validated in cell culture media, plasma, and tissue homogenates. 

The Nitric Oxide HTS Assay Kit provides a simple user-friendly, standardized and high-throughput-compatible method to quantitate the amount of NO in cell-based assays with highest sensitivity (as low as 5 pg). It utilizes a dye that reacts with intracellular NO to produce a highly-fluorescent triazole product (Ex/Em= 488/532 nm). The kit includes diphenyleneiodonium (DPI), a potent inhibitor of nitric oxide cellular production, which has been widely used to inhibit NO synthesis in order to evaluate NO function in different systems and diseases. 





Nitric oxide synthases (NOSs) are a family of enzymes that catalyze the production of nitric oxide (NO) from L-arginine and there are three isoforms of NOS: endothelial (eNOS), neuronal (nNOS), and inducible (iNOS). nNOS accounts for the production of NO in central nervous system, where NO participates in cell communication and information storage. eNOS produces NO in blood vessels and is involved with the regulation of vascular function. In contrast to other isoforms, iNOS is expressed de novo under oxidative stress conditions and produces large amounts of NO as a part of body’s defense mechanism.

PromoKine’s Intracellular Nitric Oxide Synthase (NOS) Detection Kit provides a simple, non-radiometric method for detection of intracellular NOS in the cells. It uses a dye that reacts with intracellular NO produced by NOS. The resulting fluorescence (Ex/Em = 485/530 nm) is proportional to the concentration of intracellular NOS and can be detected using a microplate reader or a fluorescence microscope.
