Histone acetylases (HATs) and Histone deacetylases (HDACs) are associated with regulation of gene transcription/expression. In general, increased levels of histone acetylation are associated with increased transcriptional activity, whereas decreased levels of acetylation are associated with repression of gene expression.

HDACs are localized in both the cytosol and nucleus and some shuttle between the nucleus and cytosol. They are transcriptional repressors with crucial roles in mammalian development and are believed to be involved in important biological activities, such as cell cycle, cell differentiation, cell proliferation, apoptosis, senescence, and epigenetic regulatory processes. Increased HDAC expression has been observed in various cancers and metastasis, while inhibition has been implicated to modulate transcription and to induce apoptosis or differentiation in cancer cells.

HDAC-3 is essential for efficient DNA replication and DNA damage control and plays also a role in cancer development, e.g. human liver cancers.
Elevated HDAC-6 activity is associated with cell motility and increases α-tubulin deacetylation, thus influencing cancer cell metastasis. In addition, mutations in HDAC-6 gene have been associated with Alzheimer's disease.Thus, HDAC-6 selective inhibitors are considered as promising targets for autoimmune, oncology and inflammatory diseases.
HDAC-8 has also been linked to number of diseases, particularly to acute myeloid leukemia, as well as parasite and viral infections.
