The GeneOFF shRNA Vectors have been developed to efficiently induce RNA interference (RNAi) by expressing small hairpin RNAs (shRNAs) in vitro or in vivo which are able to suppress distinct target genes. This is achieved by cloning an shRNA coding sequence of 45-50 nucleotides into an RNA polymerase III expression cassette. After transfection, the siRNA sequence is expressed as a fold-back stem-loop structure that is subsequently processed in the cell into functional siRNAs. The GeneOFF pPK-H1 and pPK-U6 shRNA Vectors contain the optimized U6- or H1-RNA polymerase III promoters, allowing optimal expression in a wide variety of cell types. All GeneOFF shRNA Vectors contain a neomycin resistance gene for convenient selection of stable cell lines. In addition, a variant of the pPK-H1 vector is available with a GFP reporter gene cassette to allow easy determination of the PCR fragment’s transfection efficiency.

The PromoKine GeneOFF siRNA Generation Kit provides easy and cost-effective generation of a large number of functional siRNAs from full-length target genes in vitro. The GeneOFF siRNA Genaration Kit combines rapid, high-yield synthesis of dsRNA (using T7 in vitro transcription) and extremely complete cleavage of these dsRNA templates into 22-bp siRNAs by an ultra-active form of human Recombinant Dicer Enzyme.
The kit contains sufficient reagents for generating a pool of small interfering RNAs (siRNAs) from up to 5 different genes and for 50 transfections in 24-well plates. It includes a T7 Transcription Kit for 5 transcription reactions, 50 units of Recombinant Dicer Enzyme, RNA purification columns and PromoFectin-siRNA for 50 transfections.
PromoKine also provides an effective RNase inhbitor to protect siRNA from degradation. siProtect is non-toxic to cells and very stable under a broad range of siRNA transfection conditions. It is steril and free of DNases.
