Cell Proliferation Kit I (CFSE)

PromoKine’s Cell Proliferation Kit I (CFSE) provides a convenient and sensitive way to determine cell proliferation or cell division. The assay is based on the amine-reactive dye CFDA SE (5-(and 6)-carboxyfluorescein diacetate, succinimidyl ester; also known as CFSE) which is non-fluorescent and can diffuse passively into cells. When it enters the cells, the acetate group is removed by intracellular esterases yielding the bright green-fluorescent dye carboxyfluorescein SE. Its succinimidyl ester (SE) reactive group reacts with intracellular amines and attaches the fluorescent dye to intracellular proteins, retaining it in the cells and allowing fixation with aldehyde fixatives. CFDA SE has been used as a tracer for several cell divisions as the label is stably inherited by daughter cells through successive cell divisions or after cell fusion and is not transferred to adjacent cells in a population.


Cell Proliferation/Cytotoxicity Kit II

PromoKine’s Cell Proliferation/Cytotoxicity Kit II (fluorometric) provides an easy, rapid and quantitative method to measure cell proliferation as well as cytotoxicity. The assay is based on a nuclear dye that specifically binds to nucleic acid in the cell nucleus and emits green fluorescence. The generated fluorescent intensity is directly proportional to the cell number, which can be quantified by measuring fluorescence at Ex/Em = 480/538 nm using a fluorometer or fluorescence plate reader. This assay kit provides a just add-and-read, non-radioactive, and high-throughput method that is more sensitive than MTT, XTT, or MTS-based assays, and can detect a wide linear range of 25-60,000 cells.


Cell Proliferation Kit III (EdU)

One method for detection and quantification of cell proliferation is based on the incorporation of nucleoside analogs like [3H]thymidine or 5-bromo-2’-deoxyuridine (BrdU) to into the cells' DNA during replication and visualization by autoradiography or with an anti-BrdU-antibody respectively. Both methods exhibit several limitations. Working with [3H]thymidine is troublesome because of its radioactivity and autoradiography is slow and thus not suitable for rapid high-throughput studies. The major disadvantage of BrdU staining is that the double-stranded DNA blocks the access of the anti-BrdU antibody to BrdU units. Therefore, samples have to be subjected to harsh denaturing conditions resulting in degradation of the structure of the specimen.

The PromoKine EdU-based Cell Proliferation Kits overcome these limitations, providing a superior alternative to BrdU and [3H]thymidine assays for directly measuring DNA synthesis by fluorescence microscopy, flow cytometry or fluorescence spectroscopy (e.g. using a fluorescence microwell plate Reader).
EdU (5-ethynyl-2’-deoxyuridine) is a nucleoside analog to thymidine and is incorporated into DNA during active DNA synthesis. In contrast to BrdU assays, the EdU-based Cell Proliferation Assays are not antibody based and therefore do not require harsh DNA denaturation for detection of the incorporated nucleoside. Instead, the EdU-based Cell Proliferation Assays utilize click chemistry for easily coupling fluorophores to the incorporated EdU - allowing fast and sensitive detection in a variety of dye fluorescent readouts. Furthermore, the streamlined detection protocol reduces the total number of steps and significantly decreases the total amount of time. The simple click chemistry detection procedure is completed within 30 minutes and is compatible with multiplexing for content and context-rich results.

  • Cell Proliferation Kit III (EdU-FC) for Flow Cytometry
  • Cell Proliferation Kit III (EdU-HTS) for Fluorometers/Fluorescence Microwell Plate Readers

Each kit variant is available with four different fluorescent labels (green, violet, orange, red) to enable good compatibility and flexibility with other labeling/staining approaches.
