Sirtuin (SIRT) Activity Assay & Inhibitor Screening Kits

Sirtuins are a class of proteins that possess either histone deacetylase or monoribosyltransferase activity. Sirtuins are involved in the regulation of transcription, cell cycle, apoptosis, metabolic homeostasis and oxidative stress resistance, and have been implicated in the aging process, cellular protection, as well as energy efficiency and alertness during low-caloric situations. SIRT1 in particularanti-inflammatory effects against cellular injury, and protects the cells through the regulation of mitochondrial biogenesis, autophagy, and cell metabolism in response to the cellular energy and redox status. SIRT1 also promotes vasodilation and protects vascular tissues. Activation and inhibition of SIRT1 seems to be crucial for various diseases such as cancer.

PromoKine’s fluorometric Sirtuin Activity Assay Kit provides a rapid, simple, sensitive, and reliable test to measure Sirtuin activity in a variety of samples using a fluorometer or fluorescence multiwell plate reader. In this assay, acetylated p53-AFC substrate is deacetylated by Sirtuins in the presence of NAD+ to generate the deacetylated p53-AFC substrate which is then cleaved by the developer. The released fluorescent group is detected fluorometrically at Ex/Em = 400/505 nm. The limit of quantification of the assay is
0.06 μU of recombinant human SIRT6.

The SIRT1 Inhibitor/Activator Screening Kit provides a fast, convenient, sensitive, and reliable test, which is suitable for high-throughput screening of SIRT1 inhibitors/activators. An inhibitor control (nicotinamide) is included to compare the efficacy of the test inhibitors. In this assay, Sirtuin 1 also deacetylates a fluorescently labeled substrate, followed by cleavage of the deacetylated substrate to release the fluorescent group, which is detected at Ex/Em = 400/505 nm using a fluorometer or fluorescence multiwell plate reader. In the presence of a SIRT1 inhibitor, deacetylation is impeded, preventing cleavage of the substrate and release of the fluorescent group. A SIRT1 activator however enhances SIRT1 activity resulting in a higher fluorescent signal in comparison to the control.

Analog to the SIRT1 Inhibitor/Activator Screening Kit, Sirtuin also deacetylates the substrate, which is then cleaved by the developer to release the fluorescent group whose signal can be detected at Ex/Em = 395/541 nm. In the presence of a SIRT inhibitor, the deacetylation will be impeded, preventing the cleavage of the substrate and release of the fluorescent group. An inhibitor control (nicotinamide) is included to compare the efficacy of the test inhibitors.
